The Notch signaling pathway is an evolutionarily conserved cell-cell signaling mechanism that is essential for normal embryonic development in animals. Notch proteins (Notch1-Notch4 in vertebrates) are one-way receptors activated by the Delta and Jagged/Serrate families of membrane-binding ligands. They are transported to the plasma membrane in a lytic form, but are otherwise complete polypeptides. The interaction with the ligand results in two additional proteolytic cleavages that release the Notch intracellular domain (NICD) from the plasma membrane. NICD translocates to the nucleus and forms a complex with the DNA-binding protein CSL, replacing the histone deacetylase (DAC) -corepressor (CoR) complex in CSL. Components of the activating complex, such as MAML1 and histone acetyltransferases (HATs), are recruited to the NICD-CSL complex, resulting in transcriptional activation of Notch target genes.
