GelMA Lysis Solution Instructions
Product Introduction
This product contains additives such as crude collagenase extract, which can better maintain the biological activity of the cells in the hydrogel under the premise of effectively lysing methacryloyl gelatin (GelMA) biohydrogel materials.

Storage conditions: unopened 2-8°C, valid period of Ⅰ year, mixed solution -20°C, valid period of 6 months
Instructions for use
1. Product dissolution instructions
1. Centrifuge before use, and centrifuge the powder adhered to the original tube wall to the bottom of the tube;
2. Use 2ml cell culture grade 1*PBS solution to fully dissolve the original tube powder, and filter and sterilize with 0.22um filter membrane (first add 1ml 1*PBS to dissolve, then add 1ml l×PBS to rinse the original tube);
3. The concentration of the filtrated lysate stock solution is 50mg/ml, which can be used immediately or stored at -20℃ in aliquots and protected from light.
2. Description of the method for lysing the cell-loaded GelMA gel block
1. Prepare 0.3mg/ml working solution with cell complete medium (ie 1ml working solution plus 6ul lysate stock solution); 2. After adding the working solution that can immerse the GclMA gel block into the plate well. Repeatedly blow with a pipette to separate the glue block from the bottom of the plate and break it sufficiently. The smaller the glue block, the faster the lysis speed;
3. Sterile lysis in a 37°C incubator, and observe the lysis under the microscope every 15 minutes;
4. Centrifuge at 1000 rpm for 5 minutes after full lysis. Discard the supernatant, add 5ml of complete medium and repeat the washing and centrifugation once. The obtained cells can be cultured or further analyzed for protein and nucleic acid extraction.
